Oral LSD Formulation DT120 Meets Phase III Anxiety and Depression Endpoints
New psychedelic-based treatment shows sustained symptom reduction in major clinical trials for mental disorders.
1Phase III Clinical Success
Definium Therapeutics reported that DT120, an orally disintegrating LSD formulation, successfully met primary endpoints in two phase III trials. The studies evaluated the drug's efficacy for generalized anxiety disorder and major depressive disorder. Results were presented at the Neuroscience Education Institute Fall Congress, highlighting a potential shift in psychiatric treatment paradigms.
2Anxiety Trial Performance
The Voyage study involved 214 participants with generalized anxiety disorder who received a single 100-microgram dose. Patients achieved an 11.6-point improvement on the Hamilton Anxiety Rating Scale by week 12. Statistically significant benefits appeared as early as the first week, demonstrating a 5.4-point placebo-adjusted reduction at the end of the study.
3Depression Trial Results
In the Emerge trial, 149 participants with major depressive disorder showed significant progress. The treatment led to an 8.1-point placebo-adjusted improvement on the Montgomery-Asberg Depression Rating Scale at week 6. This therapeutic benefit remained stable, with a 7.3-point placebo-adjusted improvement maintained through the 12-week mark of the clinical trial period.
4Treatment Paradigm Shift
Unlike traditional daily medications, DT120 is administered during a single supervised clinical session. Patients are monitored until discharge, which occurred within 6.4 hours on average for anxiety patients and 5.8 hours for depression patients. This model aims to replace long-term daily maintenance with a focused, one-time intervention supported by trained personnel.
5Patient Demographics Analyzed
Voyage participants had a mean age of 42, with 64% female and 36% having previously failed two or more anxiety medications. Emerge participants averaged 44 years old, with 53% having tried multiple antidepressants. These cohorts represent populations that have historically struggled to find relief through standard pharmacological interventions currently available on the market.
6Safety and Tolerability
DT120 was reported as well-tolerated across both trials, with no serious adverse events or suicidality signals observed. Most reported side effects, such as nausea, illusion, or euphoric mood, were mild to moderate and occurred primarily on the dosing day. Only one participant discontinued the study due to a treatment-emergent adverse event.
7Clinical Need Addressed
Chief medical officer Dan Karlin emphasized that current mental healthcare often requires daily pills and weeks of waiting for results. By offering a single-dose alternative, the company hopes to move beyond simple symptom management. The goal is to fundamentally alter the course of serious mental health disorders for patients suffering for years.
8Future Treatment Implications
The positive phase III data suggests that psychedelic-assisted therapy could provide a differentiated option for patients who do not respond to conventional antidepressants. Clinicians are seeking alternatives that provide substantial, sustained relief without the burden of daily adjustments. Further regulatory review will determine if this approach becomes a standard clinical practice.
